Effect of Schisandra chinensis extracts on cell apoptosis and Bcl-2 and Bax gene transcription in HepG2 cells

Objective To study the effect of Schisandrae chinensis extracts(SCEs) on the proliferation, apoptosis and Bcl-2

and Bax mRNA expression in HepG2 cells.Methods HepG2 cells were cultured and treated with different concentrations

of Schisandrae chinensis extractsSCEs,SchA,SchB and DDP.The cell growth inhibition was detected by MTT assay.Cell apoptosis was measured by 

flow cytometry.QRT-PCR was used to detect the expression of Bcl-2 and Bax mRNA.Results Treatment with SCEs,SchA,

SchB and DDP 's inhibited HepG2 cells growth in a dose-dependent manner,and the IC50 values were 41.91 μmol/L,

350.0 μmol/L,236.52 μg/mL and 17.92 μmol/L,respectively.SchA,SchB and DDP effectively induced or promoted 

HepG2 cells apoptosis.When greater or equal to 200 μg/mL,SCEs obviously promoted HepG2 cells apoptosis.mRNA 

levels of Bcl-2 and Bax were significant decreased when treated with Schisandrae chinensis extractsSCEs,SchA and SchB,but DDP increased both 

genes expression.Conclusion Schisandrae chinensis extractsSCEs can induce HepG2 cells apoptosis and reduce the expression of Bcl-2 and Bax 

mRNA,by which the mechanism still needs further research.
